Does both Node and AiMesh Router have same Wifi name, (they should do)

Your Android Phone, (S8) is it upto date and is the wifi calling using standard Android wifi calling seetings (no app)?

last question, are both Routers running the same firmware version?

For WiFi call and Skype andΒ  video issue, Could you try to disableΒ  – LAN > Switch Control > NAT Acceleration if available?

I am initially thinking that the converted DSL-AC68U is the likely cause as it is set up as Node,Β  maybe an option to make the DSL-AC68U (prime) and RT-AC68) as node. (more of a workaround, rather than fix) ;-)
